We are
committed to continuing investment in oncology research to
discover and develop more targeted and less toxic therapies than
are currently available for a broad range of cancers. We have
several different classes of compounds in various stages of
clinical development. These approaches address multiple phases
of cancer progression, including angiogenesis (new blood vessel
formation), signal transduction and programmed cell death
(apoptosis).

Advaxis is a discovery stage biotech
company developing bioengineered Listeria
monocytogenes for use as a live cancer vaccine vector.
Listeria is safe to use as a live
vaccine vector. As an environmental pathogen found in our food
supply and routinely ingested, it is a pathogen with which we
have an immune relationship. It is not laterally transmissible,
and other than the immunosurpressed and 3rd trimester
pregnant or nursing women Listeria is safe in most
people,. In the event of a problem Listeria infection
can be sterilized rapidly with a low dose of a variety of common
antibiotics.
Listeria is a pathogen that infects
the immune system and offers an unusual ability to manipulate a
number of important immune mechanisms in a simultaneous and
integrated fashion, including: stimulation of effector cells
(CD4+, CD8+, DC, macrophages, etc),
stimulation of many immune mediators (Th-1 cytokines, chemokines,
co-stimulatory molecules, GM-CSF and others), maturation of
immature immune cells and the stimulation of monopoesis, as well
as the ability to remove inhibitory influences on the immune
response (Tregs, immature myeloid cells). By infecting Antigen
Presenting Cells (APC) Listeria vaccines have the ability
to redirect a whole body immune response and focus it on which
ever tumor specific antigen is bioengineered into the
construct. By using a proprietary fusion protein in which the
microbe is engineered to secrete a tumor specific antigen fused
to a Listeria protein dramatic increases in efficacy have
been observed.
The first live Listeria vaccine in
the clinic is Lovaxin C (Lm-LLO-E7) directed against the HPV-16
E7 antigen presumed to cause most of the cervical cancer seen in
the US. In Phase I only a flu-like syndrome was associated with
Lovaxin C administration. Various studies to test Lovaxin C in
early stage disease, adjuvant models, and other protocols are
planned.
